Granisetron HCl
Indications/Uses
Prevention & treatment of acute & delayed chemo- & RT-induced nausea & vomiting (CINV & RINV); post-op nausea & vomiting (PONV).
Dosage/Direction for Use
Adult CINV & RINV prevention 1-3 mg slow IV over 30 sec or IV infusion over 5 min prior to chemo- &/or RT. CINV 1-3 mg slow IV over 30 sec or IV infusion over 5 min. Max: Not to exceed 9 mg over 24 hr. PONV prevention 1 mg slow IV over 30 sec prior to anesth induction. PONV 1 mg slow IV over 30 sec. Max: 3 mg daily in patient undergoing anesth for surgery. Childn CINV 10-40 mcg/kg IV infusion over 5 min prior to chemotherapy, may administer additional dose w/in 24 hr at least 10 min after initial dose.
Special Precautions
Cross-sensitivity to 5-HT3 antagonists eg, dolasetron, ondansetron. Signs of subacute intestinal obstruction; preexisting arrhythmias or cardiac conduction disorders, cardiac morbidities. Patients on cardiotoxic chemotherapy &/or w/ concomitant electrolyte abnormalities. Severe renal failure. Hepatic impairment. Pregnancy & lactation. Avoid use in childn <2 yr. Not to be used in neonates.
Adverse Reactions
Headache, constipation. Hepatic transaminases elevation.
Drug Interactions
Increased plasma clearance w/ phenobarb. Drugs prolonging QT interval &/or arrhythmogenic.
